Dr. Duncan’s extensive experience in the field includes applications for process monitoring and finished product inspection, notably using headspace analysis for 100 percent container closure integrity testing.

Over the years, the context of container closure integrity has broadened. It is not only linked to maintaining sterility but also to preserving critical headspace gas conditions. This is particularly relevant for certain sterile pharmaceutical products, including mRNA and live vaccines, gene therapies and products with active cells. These require ultracold storage or cryogenic temperatures to maintain stability and/or activity. Studies have indicated that ultracold storage temperatures can pose risks to traditional pharmaceutical primary packaging systems, specifically the CCI of vial-rubber stopper combinations and pre-filled syringe systems traditionally used to fill sterile pharmaceutical products.

The webinar will explore traditional container closure integrity testing (CCIT) methods and the emerging use of laser-based headspace analysis, a deterministic method that is being regularly implemented as the ‘golden tool’ for generating robust science-based CCIT data for all types of sterile pharmaceutical products.
