PathAI and Gilead Show AI-Powered Pathology Research Models Accurately Interpret Liver Histology in Patients with NASH at AASLD 2019


PathAI, a global provider of artificial intelligence (AI)-powered technology for use in pathology research, today announced the results of analyses examining the severity of nonalcoholic steatohepatitis (NASH) in liver biopsy samples from patients enrolled in the phase 3 selonsertib studies (STELLAR). Strong correlations were demonstrated between AI-powered and manual pathologist interpretations of the histological features of NASH. The data support the potential utility of machine learning models in NASH clinical trials and highlight the benefits of generating automated and quantitative assessments for staging and characterizing this disease, which affects as many as ten percent of Americans. PathAI and Gilead will present these findings at The Liver Meeting, held by the American Association for the Study of Liver Diseases from November 8-12 in Boston.

“Quantitative and reproducible assessment of liver pathology is critical for the evaluation of new therapies for NASH,” said PathAI co-founder and Chief Executive Officer Andy Beck MD, PhD. “We are thrilled to demonstrate the potential of the PathAI research platform to address this important area of drug development.”

In the first analysis (Oral Presentation #187), machine learning models were trained on liver biopsy images with more than 68,000 annotations from 75 board-certified pathologists using the PathAI research platform; images were also scored by pathologists according to the NASH Clinical Research Network (CRN) and Ishak fibrosis staging systems. The results showed that the machine learning models and the consensus of readings from the independent pathologists were in good agreement for the key histologic features of NASH. Importantly, for the staging of fibrosis, the predictions of the machine learning model were highly correlated with those of the central pathologist for both the NASH CRN (rs=0.83) and Ishak (rs=0.86) staging systems. These analyses also highlighted the potential of machine learning models to characterize fibrosis in patients with cirrhosis beyond conventional histological staging, and to do so in an automated fashion.

In the second analysis (Poster Presentation #1718), associations between the machine learning-based assessments of fibrosis, other markers of fibrosis, and the incidence of liver-related complications were evaluated in patients with cirrhosis due to NASH. The data demonstrate the heterogeneity of fibrosis in patients with cirrhosis, significant correlations between machine learning fibrosis scores and noninvasive tests of fibrosis (eg. Enhanced Liver Fibrosis [ELF] score and liver stiffness by transient elastography), and the potential to risk stratify patients with advanced fibrosis due to NASH.

“The data generated by the PathAI research platform clearly demonstrate the potential of automated, machine learning-based histologic assessment in NASH clinical trials,” said Mani Subramanian, MD PhD, Senior Vice President, Liver Diseases, Gilead Sciences. “We look forward to continued collaboration with PathAI to explore the utility of the deep learning platform to identify novel histologic features associated with treatment response and disease progression in ongoing and future trials of our NASH therapies.” Selonsertib is an investigational compound and is not approved by the U.S. Food & Drug Administration (FDA) or any other regulatory authority. The State Farm campus on Monocacy Boulevard in Frederick will soon to be buzzing with activity again thanks to Fitzgerald Realty Group’s assistance in helping Frederick Memorial Healthcare Systems (known locally as simply Frederick Health) acquire the site.

The site was valued at $38 million in January of 2018. It just closed at $48 per square foot or around $18 million, representing real value for the health care provider.

The newly named Frederick Health Village will allow Frederick Health to move its call center, home health care services and hospice, among other administrative offices to the site plus plan for further expansion in the future.

Frederick Health Hospital will not be moving from its location in downtown Frederick, but will gain some “breathing room” with the relocation of those units. Next year, the hospital plans to expand its cardiac catheter lab and upgrade both the ER and intensive care unit.

“This is a win-win for Frederick Health and the greater Frederick community,” said Jim Fitzgerald, Founder & President of Fitzgerald Realty Group who was directly involved in the negotiations. “Not only was this a smart business decision, it gives Frederick Health the opportunity to expand and refine its services to meet the growing health care needs of the local community far into the future,” he added.

Free Doggies for Dementia Photo Shoots Provide Comfort and Ease Loneliness

Photographer Carmen Davailus Buck combines pet therapy and portrait therapy to bring peace to people who have dementia and their families. “A professional portrait of a loved one gives families something special to hold on to as they cope with a devastating illness,” she said. Davailus, a Texas native, has traveled all over the country to do these free portrait sessions. She noted, “My goal is to raise awareness about Alzheimer’s and other dementia diagnoses everywhere in America.”

While portraits capture precious memories, it can be uncomfortable or unnerving for someone with Alzheimer’s Disease to sit for a photo shoot, especially with a photographer they don’t know. Carmen removes roadblocks by inviting dogs into the portrait sessions. “This allows me to capture people in natural and spontaneous situations,” said Davailus. “A dog brings out their personalities in such a beautiful way that the portraits often move families to tears. They become treasured family keepsakes.” Davailus became passionate about helping people with Alzheimer’s Disease during four decades as a nurse practitioner. She noticed many patients and their families suffered from loneliness and isolation and wanted to find a way to bring them some joy. She knows that families dealing with dementia are often financially strapped. So, she never charges for her Doggies and Dementia portraits. She noted, “The program is designed to gift the family photos and an experience that they wouldn’t normally have.”

Community Support Needed to Continue the Program: The photographer relies on corporate sponsorships to offset the costs involved. So far, she has worked with skincare company Rodan and Fields and The Legacy at Georgetown Senior Living Center. “It’s a wonderful opportunity to help an individual and to help raise awareness for Alzheimer’s Disease and related dementias,” she said.

To mark the 45th year anniversary of the discovery of the most famous hominid fossil in the world, “Lucy: The Beginnings of Humankind” is being released as an audiobook narrated by the author and her discoverer, Dr. Donald Johanson.

One of the most accomplished scholars of human origins, Johanson has produced some of the field’s most groundbreaking discoveries, including the most widely known and thoroughly studied fossil of the 20th century, the 3.2-million-year-old “Lucy” skeleton.

The 20th century was peppered with important early-human fossil finds, but Johanson’s 1974 discovery in Ethiopia that added a crucial link, prompting major revisions in our understanding of human evolution. “Lucy” possesses an intriguing mixture of ape-like features ( a projecting face and small brain) but also characteristics we consider human, such as upright walking, marking an important step on the path to Homo sapiens.

In the early 1970s the remote region of Ethiopia known as the Afar Triangle was terra incognita and its potential for illuminating our evolutionary past was unrecognized. In spite of the desolate and hostile nature of the area Johanson, with meager funding led an expedition to a site known as Hadar and began to unearth a treasure trove of fossils, including Lucy, that walked the planet some 3 to 4 million years ago. So revolutionary were these discoveries that the entire story of how we became human required substantial revision. Now some 45 years later we know more details of the vital picture of how humans came to be, but the general outlines of the human family tree proposed by Johanson’s early work continue to form the core of that story.

Johanson is committed to sharing and explaining his scientific discoveries to the general public. His efforts in this have reached across multiple media—including television, books, and his award-winning science website http://www.becominghuman.org, in addition to lecturing at universities, corporations, and public forums.
