California’s Pure Gold Forensics Will Begin Using STRmix to Interpret Mixed DNA Profiles


Pure Gold Forensics, Southern California’s only accredited, private forensic DNA laboratory, will begin using STRmix™ forensic software to resolve mixed DNA profiles in criminal investigations.

Pure Gold Forensics’ decision to use STRmix™ is based on the software’s strong track record in producing usable, interpretable, and legally admissible DNA evidence in a wide range of violent crime and sexual assault cases, as well as cold cases in which it was used to re-examine evidence originally dismissed as inconclusive.

These two probabilities are then presented as a likelihood ratio (LR), inferring the value of the findings and level of support for one proposition over the other.

STRmix™ success in accurately analyzing complex DNA mixtures has led to its use in 68 local, state, federal, and private forensic laboratories in the U.S., as well as all nine state and territory labs in Australia and New Zealand and 14 other forensic labs in countries including the U.K., Ireland, Canada, Finland, Switzerland, and Denmark.

A new version of STRmix™, Version 2.9, was recently launched. Amprion has announced the commercial rollout of its innovative SYNTap Biomarker Test, a first-in-class laboratory-developed test (LDT) for accurate and reliable detection of α-synuclein aggregates, a hallmark in various brain diseases.

The SYNTap Test aids in diagnosing synucleinopathies such as Parkinson’s, Lewy Body Dementia, and Multiple System Atrophy (MSA). In addition, the novel test helps distinguish underlying synucleinopathies in other neurodegenerative conditions, such as Alzheimer’s. Based on Amprion’s proprietary technology, Seed Aggregation Assay (SAA), the SYNTap Test identifies prion-like α-synuclein aggregates using a few drops of CSF (about one hundred microliters), confirming the presence of progressive brain disease. Test results are intended to accompany other clinical and diagnostic findings for patient case management.

“Before the SYNTap Test, there has been no specific, objective laboratory test for the diagnosis of neurodegenerative diseases such as PD, LBD, and MSA. Traditionally, patients are diagnosed based on clinical symptoms, which means their disease is relatively advanced,” said Amprion Co-founder and Chief Scientific Officer Claudio Soto, Ph.D. “Our technology detects the prion biomarkers in CSF samples years before patients show symptoms. Such early diagnosis is crucial because it enables physicians to steer their patients to proper treatments.” In addition, patients can make lifestyle changes early when their brain is healthier and more responsive to intervention. Until now, early diagnosis was not an option.

“Brain diseases affect individuals differently. For instance, ten patients can have ten different biomarker profiles,” said Dr. Russell Lebovitz, MD, Amprion CEO and co-founder. “Symptoms often overlap, leading to misdiagnosis 40 to 50% of the time. In addition, diagnosis significantly impacts drug treatment as wrongly prescribed medication can harm patients instead of helping them. The SYNTap Test provides physicians a molecular tool to greatly improve diagnostic accuracy. This is vital because personalized medicine is the key in treating neurodegenerative disorders.”

Recently awarded a US FDA Breakthrough Device Designation, Amprion’s prion detection research has been supported by the National Institutes of Health under award numbers 1R42AG058333 and 1U44NS111672, Alzheimer’s Drug Discovery Foundation (ADDF), The University of Texas McGovern Medical School, and The Michael J. Fox Foundation.

Performed exclusively at Amprion’s CLIA-registered laboratory in San Diego, California, the SYNTap Test is available throughout the US except for Maryland, New York, Pennsylvania, and Rhode Island. Market rollout in these states is expected within the next six months.

Currently, 5.8 million people are diagnosed with Alzheimer’s in the US annually, 1.4 million with LBD, and 1.5 million with Parkinson’s, adding to the millions already suffering from these diseases without a cure. And these numbers are expected to triple by 2050.

The “MVP of the Year,” a fundraising program led by PinkTie Charities in partnership with Keller Williams Core to raise a total of $250,000 to help construct two STEM classrooms at the Mariano Rivera Learning Center in New Rochelle, New York, announces the winning team of their year-long initiative; Allan Zapadinsky of Keller Williams NYC and Team Ground Breaking Ballers.

“At a time when it feels like ‘what’s in it for me?’ is a first instinct, I found myself in so many moments throughout this fundraising effort thinking ‘what else can I do for others?’” says Zapadinsky.

“Mariano, as the natural leader he is, inspired us to achieve at the highest limits, as a team, so the local kids can be their best too.”

The STEM classrooms will create an environment that expands students’ knowledge in technology, engineering, mathematics, coding and product developments. All learning center students will have the opportunity to participate in activities related to coding, UX/UI design, engineering and robotics on site or virtually. Unanimous Baseball Hall-of-Famer and five-time World Series champion, Mariano Rivera, is the ambassador of MVP of the year and leads the Mariano Rivera Foundation.

“Together, as we launch the MVP Program, we will raise much needed funds to help build our Learning Center in New York to support and stabilize the future of our students living in low income communities,” explains Mariano Rivera.

“We are grateful for organizations like Keller Williams and PinkTie and appreciate all of the MVP of the Year fundraisers and teams who have committed their time and energy to this program for the sake of the underserved minority youth. The “MVP of the Year” program consists of 10 teams all competing to see who raises the most money to help construct the two STEM Learning classrooms.

Each team ran their own fundraising events, virtual giveaways and auctions to raise a guaranteed minimum of $25,000 each. Due to last year’s COVID-19 restrictions, the 2020 winning team, “Ground Breaking Ballers,” was announced on September 23, 2021 at the Keller Williams NYC office at 99 Park Ave, New York, NY and awarded the title of “MVP of the Year,” raising nearly $40,000. All proceeds raised from the MVP of the Year fundraising program will benefit and help construct two STEM classrooms at the Mariano Rivera Learning Center by 2022.

As reported in The Guardian of Sept. 28, 2021, Greta Thunberg excoriated world politicians for climate blah blah blah and not responding to what she claims is a climate emergency, but Friends of Science Society’s upcoming event confirms that “Politicians Can’t Stop Climate Change” and Greta’s blah blah blah is misinformed.

Dr. Guus Berkhout, president of CLINTEL, the climate intelligence organization of over 900 signatory scientists and scholars who say there is no climate emergency, will present “Climate Science: Let the Data Speak” on Oct. 2, 2021, at 10 a.m. MDT as part one of Friends of Science Society’s 18th Annual Free Event online. Marc Morano, author, and climate realist activist will counter many of Greta’s claims to phase out fossil fuels ‘immediately’ in his presentation “Green New Deal: The Great Regret”, part two of Friends of Science online event on Oct. 6, 2021, at 7 p.m. MDT. Live stream link. Marc offers the public an entertaining exposé of climate hypocrisy and the muddled messages from climate catastrophe reporters. But his presentation further warns the public of the dangers of COVID lockdown becoming the method of choice for climate fanatics intent on reducing emissions. As Dr. Berkhout and colleagues show in the CLINTEL World Climate Declaration, ‘the science’ reveals there is no climate emergency, therefore there is no need to turn world energy systems upside down or strive for NetZero2050 targets.

People may wonder why the climate emergency is over, when governments around the world have embraced this theme for a few years, says Friends of Science.

The reason is found in peer-reviewed research by Roger Pielke, Jr., and Justin Ritchie, who found that ‘the science’ of climate emergency is all based on an implausible scenario known as RCP 8.5* and outdated science premises. (*Representative Concentration Pathways – 4 scenarios developed for climate research, not meant to be used for climate policy making.)

As Pielke, Jr. explains in a post entitled “The Unstoppable Momentum of Outdated Science”, this type of repetitive research error has occurred in medical research on breast cancer as well, where thousands of scientific papers were published, all based erroneously on a skin cancer cell-line.

It should be a great relief to people around the world that without reference to the RCP 8.5 scenario, there is no climate emergency and we do have time for a more thoughtful implementation of new energy technologies, says Friends of Science.

Furthermore, taxpayers will be relieved that the upcoming COP26 in Glasgow seems likely to fall apart, largely because of the attempted extortion of trillions of dollars from Western OECD nations by developing nations. Originally promised a $100/bn a year Green Climate Fund to be drawn from OECD nations as a bribe to get on board the Paris Agreement, the financial appetites of emerging nations have grown exponentially. As Canadian energy economist Robert Lyman explains in “COP26 Climate Conference in Glasgow – Show Me the Money!” Americans may be held hostage for a proposed $80 billion a year for green climate funding – a far cry from the April 2021 commitment by U.S. President Biden to increase climate aid to $5.7 billion annually by 2024!

As Friends of Science Society has always said, open, civil debate and full cost benefit analysis of climate and energy policies are essential for good public policy. Recent peer reviewed science shows that climate models overstate atmospheric warming, and the UN IPCC’s method of attributing warming to greenhouse gases is flawed, thus carbon dioxide’s warming effect on climate is no longer seen to present any imminent existential threat, consequently we do have time.

Recent study in the Journal of Urology shows PredicineCARE urine-based liquid biopsy test provides non-invasive molecular profiling for patients with bladder cancer

Predicine, Inc. announces results from a study demonstrating the concordance between tissue testing and analysis of urinary circulating tumor DNA (utDNA) while utilizing the PredicineCARE liquid biopsy NGS assay to detect somatic cancer variants in patients with urothelial bladder cancer (UBC).

The study published in the Journal of Urology evaluates the concordance between the “gold standard” of tumor tissue DNA (tDNA) with blood circulating tumor DNA (ctDNA) and urinary circulating tumor DNA (utDNA). The study is entitled “Urinary Molecular Pathology for Patients with Newly Diagnosed Urothelial Bladder Cancer.” The study utilized PredicineCARE, a 152-gene next-generation sequencing assay that evaluates somatic cancer variants.

The study found that a wide range of quantitative metrics such as cancer cell fraction, variant allele frequency, and tumor mutation burden were concordant between tDNA and utDNA, but not ctDNA. Using tDNA as the gold standard comparison, utDNA analysis achieved a specificity of 99.3%, a sensitivity of 86.7%, a positive predictive value of 67.2%, a negative predictive value of 99.8%, and a diagnostic accuracy of 99.1%. The utDNA assays outperformed ctDNA assays across all these metrics. Of note, the study found that higher levels of preoperative utDNA levels were associated with worse relapse-free survival. Finally, actionable variants, including alterations in FRGR3 and ERBB2 amplification, were identified in utDNA, which could help guide targeted therapies for patients.
