Predicting Immune Checkpoint Inhibitor Response Directly from Tumor RNA-Sequencing Data, Upcoming Webinar Hosted by Xtalks

There is an urgent need for robust predictive biomarkers for immune checkpoint inhibitor response to select patients with likely clinical benefit from this costly treatment.

Immune checkpoint inhibitors (ICI) have become the standard of care as a first- or second-line systemic treatment for patients with various cancer types. Unfortunately, many patients do not respond to this treatment and the occurrence of immune-related adverse events varies widely. There is an urgent need for robust predictive biomarkers for immune checkpoint inhibitor response to select patients with likely clinical benefit from this costly treatment.

Several biomarkers, aimed at predicting response to immune checkpoint inhibitors, have been proposed, including PD-L1 expression, tumor mutation burden (TMB), infiltration of cytotoxic T-cells in the tumor, Microsatellite Instability (MSI) and expression of various immune gene signatures. However, these individual biomarkers have suboptimal performance and multiple complementary omics technologies are required to properly quantify them. Integration of these multi-omic biomarkers has proven valuable in increasing the accuracy and robustness of immune checkpoint inhibitor response prediction.

The featured speakers have developed a computational pipeline that determines the expressed mutation burden (eTMB), MSI status, fraction of infiltrating immune cells and various immune gene expression signatures directly from the RNA-sequencing profile of the tumor. Each biomarker is quantified using a dedicated algorithm that applies machine learning (eTMB and MSI) or computational deconvolution (infiltrating immune cells) and integrates external data sources to maximize performance. Algorithm performance has been validated on large cohorts of tumor RNA-sequencing data with matching gold standard quantification of said biomarkers. Moreover, integrating these biomarkers improves prediction of response to checkpoint inhibition therapy.

Register for this webinar to learn how this computational approach enables the quantification of various immune checkpoint inhibitor response biomarkers from a single omics layer (RNA-seq) and improves response prediction.

Join Pieter Mestdagh, Computational Biology Manager, CellCarta, for the live webinar on Wednesday, May 31, 2023, at 11am EDT (4pm BST/UK).

empty tomb®’s Mission Match® Revised Focus Builds on Work Begun in 2002

Mission Match’s focus on under-age-five child survival builds on more general mission work done by churches from 2002 through 2016 with matching contributions.

empty tomb, inc.’s Mission Match’s focus is now on churches helping, in Jesus’ name, to meet the goals to reduce the rate of under-age-five child deaths in 40 countries.

These 40 countries, as of 2015, were behind the curve in meeting goals set by world leaders to reduce the rate of child deaths.

A church in the U.S. can apply for a matching contribution from $500 to $3,000 in $500 increments.

The church mission projects can, for example, support medical workers, provide a specific item needed at a medical facility, or furnish supplies that contribute to child survival. It is understood that other age groups might also be assisted by the projects.

Mission Match is seeking 30 churches to launch a next phase in developing this mission incentive tool. Churches can apply for matching contributions for their medical mission projects.

Mission Match’s focus on under-age-five child survival builds on more general mission work done by churches from 2002 through 2016 with matching contributions.

From 2002 to 2016, churches could apply for Mission Match matching contributions for any type of mission project the congregation chose. A total of 86 churches from 23 states carried out 112 projects during those years. Mission Match provided $222,000 that was matched by a reported $335,115 raised in the congregations for their projects.

Medical Marijuana Treatment Clinics Opens as TruReleaf MD in Jackson, Mississippi

“This new industry promises to offer us natural alternatives to powerful narcotics, which have caused senseless deaths of too many of our neighbors and their loved ones.” says Dr. Roberts, licensed medical marijuana physician.

Jackson — TruReleaf MD is expanding to Jackson, MS, as one of the state’s first medical marijuana physicians providing relief for patients through medical cannabis certifications with in-person reviews of medical conditions and medical records.

With the passage of Senate Bill 2095, Mississippi’s Medical Marijuana Program was streamlined, providing expanded medical marijuana care to patients. One of the significant provisions allows physicians to recommend medicine to patients if they believe that their patient’s qualifying debilitating medical condition would benefit from medical marijuana or that it would alleviate the symptoms of a qualifying debilitating medical condition.

TruReleaf MD comes to the state with over seven years of experience providing compassionate care in Florida as MMTCFL. Following the legalization of medical marijuana for qualifying conditions, the Medical Marijuana Treatment Clinics of Florida quickly became a pillar in the medical marijuana industry, redefining what it means to be a medical marijuana patient and providing a natural relief option to those seeking care.

With this experience, TruReleaf MD aims to restore patient quality of life through cannabis-based alternative treatments. They believe that relief is fundamental to having control over one’s life and aim to give patients the necessary tools to make the best choice for their own health.

“This new industry promises to offer us natural alternatives to powerful narcotics, which have caused senseless deaths of too many of our neighbors and their loved ones.” says Dr. Roberts, licensed medical marijuana physician. The clinic will allow patients more opportunities to take control of their conditions, putting them one step closer to creating the life they want.

Using Machine Learning to Find High Potential Lead Molecules, Upcoming Webinar Hosted by Xtalks

Using a rapid combinatorial library and microfluidic high-throughput screening technology, it is possible to quickly design, build and screen molecular libraries to discover bioactive molecules (and inactive molecules) with quality dose-response data.

The promise of machine learning to dramatically reduce timelines in a lengthy drug discovery process is exciting, but it relies on the ability to generate a trusted set of empirical data and the continuous iteration of models.

In this webinar, the featured speakers will demonstrate their iterative and machine learning-driven discovery engine across multiple disease targets to predict novel, active chemical scaffolds and recognize SARs otherwise not identified by public data alone. Using a rapid combinatorial library and microfluidic high-throughput screening technology, it is possible to quickly design, build and screen molecular libraries to discover bioactive molecules (and inactive molecules) with quality dose-response data. This dose-response data allows library-wide SAR characterizations, empowering deep-learning QSAR model predictions and novel lead generation. The result is core starting points and activity-predictive models that expedite decision-making steps during chemical series progression.

The frontier of medicine is often fraught with early discovery decisions based on limited empirical data. While biologics discovery has been accelerated by leveraging nature’s tools for site-selective mutagenesis and rapid maturation or optimization, small molecule discovery is in desperate need of new tools that will similarly allow rapid learning cycles and optimization of clinically relevant drug properties.

In this webinar, the speakers will explore how empirical and computational platforms scout chemical space, generate measures of SAR and map the critical structural patterns to predict activity and guide development. They also explore the molecular space not previously examined and create predictions to streamline optimization toward higher potential lead molecules.

Join this webinar to learn how custom chemistry and machine learning bring medicines to patients faster.

Join experts from 1859, Ghotas Evindar, PhD, SVP Discovery; Andrew MacConnell, PhD, Cofounder and Scientific Fellow; and Hossam Ashtawy, PhD, Director of AI and ML, for the live webinar on Thursday, May 25, 2023, at 11am EDT (4pm BST/UK).

Ideal Option Opens Addiction Medicine Clinic in El Dorado

Ideal Option, a national leader in evidence-based outpatient treatment for addiction to fentanyl, heroin, methamphetamine, alcohol and polysubstance, is now welcoming new patients in El Dorado. The new clinic, located at 625 E Main St, will provide primarily buprenorphine-based medication-assisted treatment with an onsite addiction medicine specialist.

While opioid prescribing and heroin use is on the decline, overdose deaths continue to increase in Arkansas, largely due to the prevalence of fentanyl in the illicit drug supply.

According to the Arkansas Take Back Opioid Dashboard, overdose deaths in Union County increased by 50% and narcotic arrests increased by 39% from 2020 to 2021.

Fentanyl is regularly mixed with other illicit drugs to make them more addictive, often without the user knowing. Polysubstance use disorder is therefore now the most common diagnosis at Ideal Option. According to their Annual Patient Outcomes Report for Arkansas, in 2022, over half of all Ideal Option patients tested positive for more than one substance at enrollment, and 24% tested positive for three or more substances.

The report does offer hope, however, by showing how effective medication-assisted treatment is for reducing isolated and polysubstance use. For patients who remained in treatment with Ideal Option during 2022, lab testing shows:

  • 87% less total opioid use
  • 94% less fentanyl use
  • 96% less heroin use
  • 77% less methamphetamine use
  • 90% less cocaine use

The new clinic in El Dorado is Ideal Option’s tenth location in Arkansas and is opening at a time of desperate need for expanded access to evidence-based treatment from experienced providers. Ideal Option has been specializing in addiction medicine using buprenorphine-based medications such as Suboxone® for 11 years and currently operates 90 clinics in 11 states.

“We continue to see a concerning rise in fentanyl use across Arkansas, creating an urgent demand for specialized treatment,” said Aaron Bunch, community outreach coordinator for Ideal Option. “I am excited to get to work in the great community of El Dorado to tackle this worsening crisis.”

The new Ideal Option clinic in El Dorado is open Monday – Thursday, 7:30 a.m. – 6 p.m. and is located at 625 E Main St, El Dorado, AR 71730.

Most forms of insurance are accepted, including Medicare and Medicaid, and patients can typically get started on treatment for fentanyl, heroin, alcohol or other substances within 1-3 business days.

Patients can make an appointment by calling 1.877.522.1275 or visiting http://www.idealoption.com.

About Ideal Option

Headquartered in Kennewick, Washington, Ideal Option was founded in 2012 and has since helped more than 65,000 patients through a network of 90 office-based medication-assisted treatment (MAT) clinics across 11 states. With a mission to serve under-served communities, Ideal Option accepts most forms of insurance including Medicaid and Medicare. Financial assistance and payment plans are also available.

Ideal Option’s team of medical providers carry certifications in Addiction and Emergency Medicine, Internal, and Family Medicine, among other specialties. The company also employs social workers, caseworkers, counselors, and mental health practitioners. This holistic approach helps drive positive outcomes, including family stability, stable housing, improved overall health, and reduced rates of recidivism.

In all the communities it serves, Ideal Option collaborates with existing stakeholders and providers to improve the continuum of care. This approach includes partnerships with emergency rooms and county jail systems, where individuals with substance use disorder often appear, as well as collaborating with numerous support agencies and municipal programs addressing social needs such as housing, mental health, and nutrition support.

Pastor Shares Her Story of Discovering Her Sexuality and Her Authentic Self

News Image

Growing up under the influence of her family’s religious teaching and beliefs of the Catholic Church, Reverend Lisa R. Kelson spent a large portion of her life succumbing to their patriarchal teachings and ignoring her own inner compass. In her new book, “A Lesbian’s Awakening to Purpose, Self-Love, and God: A Soul’s Journey to Self-Awareness, Identity, and Truth,” she shares her personal journey to spiritual enlightenment and coming to terms with her authentic self.

“It has taken me nearly 60 years to wake up to my true authentic self as a lesbian,” Kelson said. “In my younger days, I had no idea that there was a deeper and higher self, waiting to be discovered. For many years, I lived from my lower aspect, while trying to fit into society’s social rules and norms.”

In “A Lesbian’s Awakening to Purpose, Self-Love, and God,” Kelson uses her personal experience as an example of how readers can break free of their preconceived ideals of who they are expected to be, and instead free their souls to be their true selves. She addresses the harm that can come from religious teachings, and how it can be exclusionary to those the church does not deem “perfect.”

“When one begins breaking down scripture, outside the oppressive lens of patriarchy,” Kelson said, “one can finally understand how scripture is misunderstood in our current age. These writings were written in an oppressive era, and the literal interpretation are not in alignment with deeper truths.”

Kelson doesn’t want readers to rely on anyone else’s truth, instead she wants them to discover their own.

“My hope in sharing my awakening journey,” Kelson said, “is to help those who feel lost and out of sorts with life. This journey is spiritual and not religious. The answers are already within, waiting to be uncovered.”

“A Lesbian’s Awakening to Purpose, Self-Love, and God: A Soul’s Journey to Self-Awareness, Identity, and Truth”

About the author

Reverend Lisa R. Kelson is a native Floridian. After completing her Bachelors of Sport’s Science Education, she enlisted in the Army. She completed her service after returning home from the Gulf War. Upon the completion of her master’s degree in technology education from the University of Georgia, she returned home to Florida. She soon realized that God was calling her to ministry, and she enrolled in Episcopal Divinity School where she completed her Master of Divinity. While this experience prepared her for church ministry, she would soon awaken to a higher spiritual path where she shifted her focus to awakening her higher self. This path led her to understanding deeper truths, that led to her awakening and freeing her soul. To learn more, please visit https://www.reverendlisarkelson.com/.
